Abstract: Immunogenic cell death (ICD) enhances tumor immunogenicity through the spatiotemporally regulated exposure or release of damage-associated molecular patterns (DAMPs), among which cell surface exposure of calreticulin (CRT) represents a critical early event. Accumulating evidence indicates that reactive oxygen species (ROS), endoplasmic reticulum (ER) stress, and the unfolded protein response (UPR) form a mutually amplifying signaling network that plays a central role in regulating CRT translocation and ICD induction. However, current studies remain largely correlative, and the key regulatory nodes, temporal dynamics, and rate-limiting steps have yet to be fully elucidated. This review focuses on the ROS–ER stress–CRT signaling pathway, systematically summarizing its molecular mechanisms and dynamic features, and highlighting current advances and critical gaps in oral and salivary gland tumors. In particular, potential research directions are proposed for mucoepidermoid carcinoma, where mechanistic evidence remains limited. Elucidation of this regulatory network may provide a theoretical basis for enhancing antitumor immunity and optimizing combination therapeutic strategies.
